The MBioSEQ Ridom Typer Server does not require an internet connection. However, it is strongly recommended that the MBioSEQ Ridom Typer Client is operated on a computer with an active internet connection, as some features listed below are not available in offline mode. If operating the client without an internet connection is unavoidable, the following steps can be used to start Ridom Typer. Please note that these steps apply only to the Windows version only. The Linux version of MBioSeq Ridom Typer always requires an active internet connection because it must download bioinfomatic tools from anaconda.org.

Limitations working in Offline mode

  • Public cgMLST / Accessory task templates with complex types (CTs) and public nomenclature are unavailable.
  • MLST task templates are only available with schemes and alleles up to 31/12/2024 (as per PubMLST terms and conditions])
  • The SARS-CoV-2 Pangolin task template cannot be used.
  • The S. aureus spa-typing task template cannot be used.
  • ENA upload functionality is unavailable.
  • NCBI download functionality for genomes and raw reads cannot be used.
  • Geocoding functionality in comparison table is unavailable.

Steps to start Ridom Typer in Offline mode

Activating the License

Direct license activation will not work without an internet connection. In this case, a dialog box will appear prompting you to save the activation request as an XML file. Use the button 'Save Activation Request' to save the file and send it to support@ridom.de via email.

You will then receive an activated license file that can be imported by using the 'Import License File' option in the License Activation dialog.

Installing Windows Subsystem For Linux

Installing a downloaded WSL image

If no internet connection is available, the image for the Windows Subsystem For Linux installation cannot be downloaded automatically. Therefore, the Linux image file must first be downloaded manually from https://www.ridom.de/download/wsl/ridom_ubuntu_112.0.tar.gz using a computer with an internet connection, and then transferred to the computer on which the client is installed. Next, WSL must be one-time enabled (admin right are required). Finally, the Linux distribution with all required tools must be installed. Start the MBioSEQ Ridom Typer client and invoke in the Help menu Tux.png Linux Distribution for WSL. To install the above downloaded file, select the Install from File command in the Source of the dialog. After the import is finished a system check will be run. If all checks succeed with OK, WSL was successfully installed and all tools are working.

Downloading and Importing Offline Task Templates

If there is no internet connection available, the 'Download & Add' function in the project editor will not work. In this case, the task templates must be downloaded manually from https://cgmlst.org/offline and added to the project manually using the 'Add' and 'Import from File' buttons.

The task templates can be downloaded all at once as a zip archive (with button 'Download All Task Templates'; once per week updated) or separately from the species-specific pages. The species-specific pages also show task templates that work for several bacteria (e.g., NCBI AMRFinderPlus). You only need to download and import these generic task templates once.

The cgMLST/Accessory task templates that are listed there use a local allele nomenclature and do not have a CT. The MLST task templates that can be downloaded contain only alleles up to 31/12/2024, as newer alleles require registration at PubMLST.org. If newer MLST alleles are required, the alleles/profiles must be manually downloaded after registration and manually added into a new task template.
